Wood samples taken from the woody layer are examined under a microscope.

Some species have identical microscopic characteristics, in which case the test will stop at determining the family of the wood species. DNA analysis can be used to supplement this species identification.

Fungi, insects and their characteristic features are observed under a magnifying glass and/or microscope in order to identify them.

Fungal DNA analysis (lignivorous fungi and/or moulds) is generally performed when the fungal elements are in too poor a condition or too insignificant to allow identification by observation techniques. DNA extraction, followed by amplification with specific primers, and sometimes sequencing, can enable identification.

Indoor air quality analysis is carried out to determine the amount of mould and to identify the mould in contaminated homes.
